1. Use curtains to block light, turn off room lights, remove the dust cover from the microscope, ensure good ventilation of the microscope lamp housing and no covering. 2. Insert the power plug of the mercury lamp housing into the socket on the back of the fluorescence power supply, then insert the plug of the fluorescence power supply into a 220V external power source.
3. Turn on the power switch, the voltmeter shows the power supply voltage. If the power supply voltage fluctuation is not greater than ±5% of the rated voltage value, press the start switch to ignite the mercury lamp. If the mercury lamp fails to ignite at once due to cold weather or voltage instability, press the switch several times. After the ultra-high pressure mercury lamp arc reaches a stable state and achieves maximum luminous efficiency, work can begin.
4. Centering the bulb:
(1) Place any specimen on the stage;
(2) Rotate the condenser knob on the microscope arm to move the condenser out of the light path, rotate the filter set changeover wheel to bring the violet (V) or blue (B) or green (G) excitation filter set into the light path, and bring the 10X fluorescence objective into the light path;
(3) Adjust the coarse and fine focus knobs to bring the specimen image into clear focus;
(4) Push the condenser focusing rod on the right side of the vertical illuminator forward and backward to bring the field diaphragm image into sharp focus, rotate the field diaphragm lever to reduce the field diaphragm, adjust the field diaphragm centering screws to center the field diaphragm, and then open the field diaphragm to its maximum;
(5) Rotate the condenser knob on the microscope arm to move the condenser into the light path, adjust the condenser rod on the lamp housing forward and backward to bring the mercury lamp arc image into sharp focus within the field of view;
(6) Adjust the bulb horizontal and vertical adjustment screws on the lamp housing to center the mercury lamp arc;
(7) Adjust the reflector horizontal and vertical adjustment screws to separate the reflected image of the light source from the mercury lamp arc;
(8) Rotate the condenser knob to move the condenser out of the light path; at this point, the field illumination is uniform.
